Cutting Edge Cutting Edge: Mucosal Application of a Lyophilized Viral Vector Vaccine Confers Systemic and Protective Immunity toward Intracellular Pathogens

چکیده
A major problem of current vaccines is storage stability, often requiring strict maintenance of cold chains. In the course of the eradication of smallpox, a freezedried vaccinia virus (Dryvax), which proved to be very stable, was used to overcome this limitation. However, Dryvax needs to be reconstituted before usage and is administered using a bifurcated needle, procedures that pose a number of additional health risks. We report in this study that a stable, lyophilized, modified vaccinia virus Ankara (MVA) vaccine can be directly applied to the nostrils of mice without previous reconstitution. This direct mucosal application induced systemic Ab and T cell responses comparable to those achieved by i.m. administration. Importantly, mucosal application of lyophilized MVA induced long-lasting protective immunity against lethal bacterial and viral challenges. These data clearly demonstrate the potency of a simple needle-free vaccination, combining the advantages of mucosal application with the stability and efficiency of lyophilized MVA. متن کاملExperimental Study of the Effect of Laser-Cutting Process Parameters on Heat Distribution and Cutting Edge Quality of Steel Sheets
This study aimed to evaluate Laser-cutting process of Mild Steel St37 sheets with two different thicknesses. In order to evaluate the quality of cutting edge and the depth of the area affected by the heat, several tests were conducted to determine the effect of parameters such as Laser power, Laser beam movement speed and pressure of axillary gas on the quality of cutting edge and the depth of ...
متن کاملCutting Edge Cutting Edge: Lung Mucosal Th17-Mediated Responses Induce Polymeric Ig Receptor Expression by the Airway Epithelium and Elevate Secretory IgA Levels
Polymeric Ig receptor (pIgR) is a central player in mucosal immunity that mediates the delivery of polymeric IgA and IgM to the apical surface of epithelial cells via transcytosis. Emerging evidence suggests that Th17 cells not only mediate autoimmunity but also play key roles in mucosal host defense against pathogens. We demonstrate that OVA-specific CD4 Th17 cells, in addition to causing neut...
متن کاملGastric invasion by Trypanosoma cruzi and induction of protective mucosal immune responses.
Trypanosoma cruzi is an intracellular parasite transmitted from a reduviid insect vector to humans by exposure of mucosal surfaces to infected insect excreta. We have used an oral challenge murine model that mimics vector-borne transmission to study T. cruzi mucosal infection. Although gastric secretions have microbicidal activity against most infectious pathogens, we demonstrate that T. cruzi ...
متن کاملAn Instantaneous Rigid Force Model For 3-Axis Ball-End Milling Of Sculptured Surfaces
An instantaneous rigid force model for prediction of cutting forces in ball-end milling of sculptured surfaces is presented in this paper. A commercially available geometric engine is used to represent the cutting edge, cutter and updated part geometries. The cutter used in this work is an insert type ball-end mill. Intersecting an inclined plane with the cutter ball nose generates the cutting...
